From the answer, I now know there is a manufacturing defect with si1152. My question is, do I need to change my schematic and use the LED3 pin to drive the other led and pull up the LED2 pin,
or
use the same LED2 pin but write code to drive LED3 (enable LED3 and set current setting for LED3, instead of LED2) ?

You should change your schematic to drive the LED3 pin instead. The software code should enable LED3 and set LED3 current.
SI1152 LED2: No signal on LED2 pin.
I am using SI1152 to drive two LEDs to take two different measurements respectively. The LED connected to LED2 pin is not receiving any signal. I came across this question https://www.silabs.com/community/sensors/forum.topic.html/si1152_cannot_driveled2-dN3m.

You should change your schematic to drive the LED3 pin instead. The software code should enable LED3 and set LED3 current.
